Does anybody know if modded/rooted devices will get OTA updates? I'm running CyanogenMod 4.0.1 and am curious to see what happens.

From my understanding of the issue is that, you will no longer recieve access to the Google Cloud/Experience when Donut rolls out...

I had my G1 running the same software, not wanting to lose my connection I unwillingly went back to 1.5 on the cyanogen mod website, www.cyanogenmod.com well you can read for yourself.

Of course rooted phones won't get the update! Think about it. Your root access would vanish. However, if you are running Cyanogen's mod, you can download an updater app from the market that will allow you to download and apply 1.6 OTA-ish.

If you have a rooted phone you could have had 1.6 weeks ago. I've had 1.6 for a while now. Check xda-developers there are 1.6 roms out there. At this point cyanogen is working on a legal 1.6 because he got a C&D from google. Check in a few days at xda.
